Description | EliKine™ Human Leptin ELISA Kit has high sensitivity and excellent specificity for detection of Human Leptin . No significant cross-reactivity or interference between Human Leptin and analogues was observed. LEP gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in LEP gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. LEP gene has also been linked to type 2 diabetes mellitus development. EliKine™ Human Leptin ELISA Kit employs a two-site sandwich ELISA to quantitate Leptin in samples. An antibody specific for Leptin has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Leptin present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Leptin is added to the wells. After washing, proprietary EliKine™ Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Leptin bound in the initial step. The color development is stopped and the intensity of the color is measured. |
